Organic synthesis

MEDSynth employs state-of-the-art techniques such as parallel chemistry, solid phase and microwave-assisted organic synthesis (MAOs), and classical solution-phase organic synthesis to increase the outputs. The MEDSynth labs are fully equipped for both liquid and solid-phase syntheses.
Specifically :

  • Teledyne Isco CombiFlash System
  • Medium Pressure Liquid Chromatography (MPLC Buchi 688)
  • High-pressure PARR® reactor (high-pressure reactions)
  • Kugelrohr distillation apparatus
  • Analytical HPLC (Merck-Hitachi L-6200, double pump system, UV detector, autosampler)
  • Analytical Perkin Elmer UHPLC FLEXAR FX-15 Platform
  •  UltiMateTM 3000 Semi-Preparative System
  • 500 mL Reactor-Ready Controlled Lab reactor (temperature range: -70°C/+230°C; pressure range: 10 mbar-0.05 bar; max rate: 500 rpm; software: AVA) Radleys UK

Our group have access to other facilities at the Department of Drug Science and Technology (University of Torino), due to internal collaborations:

  • 600 MHz Jeol NMR
  • Milestone MicroSYNTH microwave oven (two units)
  • Milestone SinthWave microwave oven
  • Waters Binary pump 1525EF, 2998 Diode array detector, 2424 ELS detector, WFC III fraction collector.
  • G.E. Akta Purifier 10/100 System
  • Waters FractionLink Autopurification System (Binary pump 2525, 2767 Autosampler/fraction collector, 2996 Diode array detector, ZQ ESCi- MS detector)
  • Agilent 6850 GC System equipped with 5973 Mass Selective Detector

ADME & Drug-like properties

Once the compounds have been synthesized, the ADME facility is responsible for the following profilation:

  • Chemical-physical properties of the new compounds, in particular thermodynamic solubility and LogD measurement
  • Evaluation of the stability of the compounds in different media (PBS, SGF, SIF, serum)
  • Investigations of microsomal metabolism and stability in S9 fractions
  • Serum protein binding
  • Evaluation of in vitro enzyme-inhibiting activity

To perform our analysis, the ADME laboratory is equipped with:
  • Analytical Perkin Elmer UHPLC FLEXAR FX-15 Platform
  • UV-visible spectrophotometer VARIAN 50 BIO
  • Thermomixer Shaker BIOER MB-102
  • Centrifuge ALC PK 110
  • Refrigerated Microlitre Centrifuge HERMLE Z216MK
  • Incubator NUVE EN032

In silico studies

MEDSynth group is also able to perform in silico drug design through the use of different devices.

  • Six Oculus META QUEST 2 supporting Nanome software for the immersive studies of small molecule-protein interactions.
  • Centos 7 molecular Docking station: GOLD, Maestro, HADDOCK.
  • All our computational systems are supported by the use of MolBook UNIPI software for chemical data management.
